OPTIMIZING DNA EXTRACTION WITHOUT LYOPHILIZATION OR LIQUID NITROGEN GRINDINGFROM AFRICAN YAM BEAN (SPHENOSTYLIS STENOCARPA HOCHST EX. A. RICH.) HARMS TISSUES

DNA extraction is the first committed step in every molecular biological endeavour. This biomolecule can be isolated from any material for subsequent downstream processes. Most available protocols nucleic acid entail use of either lyophilized tissues or liquid nitrogen tissue grinding. These procedures pose a major limitation such protocols, especially parts world where and freeze-dryer are not only expensive, but also very difficult to procure. The current study was an attempt optimize procedure African yam bean ({Sphenostylis stenocarpa Hochst ex. A. Rich.}Harms) tissues, employing sodium dodecyl sulphate (SDS)-phenol method,without lyophilization grinding.The effect grinding leaf with washed sand protocol, phenol extraction, precipitating temperatures, ammonium acetate washing on purity yield were investigated. Spectrophotometric analysis used determine extracts. results showed that plant acid-washed precipitation at -20 oC significantly improved single treatment sufficient obtaining pure extract. Comparatively, preferred root stem extraction. findings this indicated high quality could extracted without costly lyophilization.
